NIR Protein Analysis

What NIR measures when it reports protein, where it works well, where it fails, and how it compares with the Kjeldahl reference method in routine QC.

The short answerNIR estimates protein from how a sample absorbs near-infrared light, using a calibration built against a reference method such as Kjeldahl or Dumas. In whole wheat it can land within about 0.1% of Kjeldahl, but only as well as its calibration and reference data allow.

What NIR actually measures

NIR does not count nitrogen. It records how the sample absorbs light across the near-infrared range and a calibration model turns that spectrum into a protein value.

The model is only as good as the reference values it was trained on. Kjeldahl is the AOAC reference for nitrogen to crude protein (acid digestion, distillation and titration). Dumas combustion is the modern alternative.

Where it works

Grain elevators use it to bin and segregate by protein within a minute per sample. Flour mills use it to control wheat protein, moisture and ash. Feed mills run it on incoming ingredients because soybean meal protein varies from 44% to 49% depending on origin and crush plant. Dairy plants use it for raw milk and powder.

Where it fails

Accuracy is bounded by the reference method. If your Kjeldahl values scatter by 0.3% protein, no instrument can beat that. Unrepresented samples, particle size changes and calibration drift cause most bad readings.

Questions

How accurate is NIR for wheat protein compared to the lab?

NIR analyzers can reach crude protein results within about 0.1% of the reference method (Kjeldahl) in whole wheat. That is enough for binning, segregation and contract decisions, but it depends on a robust calibration.

Does NIR replace Kjeldahl?

For routine screening, often yes. Kjeldahl or Dumas still anchor the calibration and settle disputes. NIR is only as accurate as the reference data used to build it.

Why does my NIR protein drift?

Common causes are new sample types not covered by the calibration, particle size changes, and instrument drift. A slope and bias adjustment fixes a constant offset; a trend means recalibrate.
